    Vibration-reduced anxiety-like behavior relies on ameliorating abnormalities of the somatosensory cortex and medial prefrontal cortex

    Although PV INs are also expressed in SSC, playing critical role in feedforward and feedback synaptic inhibition onto a diverse set of cell types, such as pyramidal cells, other type interneurons, and themselves (Naskar et al., 2021), how RS and SD impact on it remained unclear. PV INs are characterized by a dense local axon and dendritic arbors (Tremblay et al., 2016). Therefore, we next carried out immunohistochemical staining to examine the number of PV INs in both the SSC and mPFC. Our findings showed a considerable reduction in the number of PV INs in the SSC of both RS (F(1,59) = 8.830, P < 0.001, P < 0.001; Ctrl vs. RS) and SD (P < 0.001; Ctrl vs. SD) mice. However, this reduction was significantly less in the RS and SD mice that received TSB-vibration treatment (RS vs. vibration + RS: P = 0.009; SD vs. vibration + SD: P = 0.011; Figure 7A and B). In contrast, the number of CaMKII-positive cells remained unchanged among groups (F(1,59) = 1.015, P = 0.417; Figure 7C).
    We also analyzed the number of PV INs in the mPFC. As in the SSC, numbers were reduced in RS (F(1,59) = 4.971, P < 0.001, P = 0.029; Ctrl vs. RS) and SD mice (P = 0.021; Ctrl vs. SD), and TSB vibrations prevented this reduction (RS vs. vibration + RS: P = 0.045; SD vs. vibration + SD: P = 0.043; Figure 7D and E). Consistent with our western blotting results, the immunohistochemistry for CaMKII showed an increased number of CaMKII-positive cells in RS (F(1,59) = 6.074, P < 0.001, P = 0.003; Ctrl vs. RS) and SD (P = 0.019; Ctrl vs. SD) mice, and a normal number of cells after vibration treatment (RS vs. vibration + RS: P = 0.018; SD vs. vibration + SD: P = 0.044; Figure 7D and F).
