To further validate the changes in Spi1 expression and determine its subcellular localization, mice were injected with collagenase and sacrificed 72 h later for immunofluorescence staining (n = 6). Consistent with the above results, Spi1 was expressed at low levels in normal brains (Figure 3A and Additional Figures 7–9). Conversely, the Spi1 signals colocalized with Iba1-positive cells in the perihematomal area and dramatically increased after ICH (Figure 3A and B). These results suggest that Spi1 regulates microglial/macrophage activation and infiltration following ICH.
