摘要:

 

目前得到的公识的是严重的脊髓损伤会导致永久性瘫痪。最近有研究者提出一项脊髓融合研究方案,其中涉及的脊髓损伤是急性可控、锐器横断伤,这种损伤并不是临床中最常见的,临床中大多数脊髓损伤是由于机械性作用、出血、瘢痕和囊肿所致。早在1963年神经外科医生Freeman就在动物实验中观察到脊髓横断伤后数月,损伤动物的运动功能会缓慢恢复,更重要的是银染结果显示,大量正在生长的轴突连接了断端,且这种恢复可在融合剂的作用下加速。我们用手术刀片切断小鼠颈髓,然后用细胞膜融合剂PEG对脊髓断面进行融合。术后4周小鼠运动功能出现恢复,然后应用免疫荧光染色结合透射电镜对小鼠损伤颈髓融合部位进行观察,发现有神经发芽现象。发芽神经元主要定位于lamina VIII层,该层是小鼠颈髓固有神经元聚集之地。
