摘要: Neural Regen Res:铁与溶酶体系统及神经炎症的关系
铁积累和神经炎症是许多神经退行性疾病的主要特征。但是,致病机理的层次结构以及这些过程之间的分子关系仍不清楚。
来自智利迭戈波塔莱斯大学的Daniel A. Bórquez团队认为在亚细胞水平精心调节大脑中存在的几种细胞类型之间的铁含量及其分布,可以满足对铁的需求,同时还能避免铁的毒性。神经退行性疾病患者大脑中的小胶质细胞可以通过一种非生物可利用的形式隔离铁,引发铁蓄积和功能性铁缺乏症的悖论性条件,从而对溶酶体功能产生负面影响。

Abstract: Iron accumulation and neuroinflammation are cardinal features of many neurodegenerative diseases (Urrutia et al., 2021). However, the hierarchy in the pathogenic mechanisms and the molecular connections between these processes are still obscure. In this perspective, we will discuss current evidence showing that aging, the main risk factor for neurodegenerative diseases, negatively impacts lysosomal function by triggering a paradoxical condition of iron accumulation accompanied by functional iron deficiency. This condition is self-sustaining in the neuroinflammatory process.
