Abstract: Autophagy is well-known for delivering cargo materials to lysosomes for proteolytic digestion. Recently, autophagy has emerged as a key mechanism in unconventional protein secretion (UPS). This perspective introduces unconventional secretion pathways, focusing on secretory autophagy and its role in secreting protein aggregates associated with neurodegenerative disorders. We also explore additional neuronal functions of secretory autophagy beyond the release of protein aggregates. We propose autophagosomes as transport organelles that deliver cargo material directly from the endoplasmatic reticulum (ER) to the plasma membrane rather than solely to lysosomes.
