摘要:

携带Atoh1的腺病毒可以感染耳蜗柯替氏器,并在支持细胞区出现类似毛细胞的表面标准物,但腺病毒携带的基因表达属于瞬时表达,这无疑限制了其临床应用,而慢病毒是一种可以稳定、持续表达目的基因的载体。实验以携带Atoh1的慢病毒通过圆窗途径导入大鼠耳蜗30d后,大鼠耳蜗内的结构及听功能无明显影响,且携带的Atoh1基因使耳蜗支持细胞区生成毛细胞样细胞,并表达毛细胞表面标志物Myosin7a。说明慢病毒对耳蜗毛细胞和听功能没有明显危害,慢病毒携带的目的基因可持久感染正常大鼠耳蜗,作用安全有效。
