韩国首尔国立大学兽医科学研究所的Dae Young Yoo等最新一项研究证实,海马CA1区锥体细胞层铁过载在短暂性脑缺血后12h最为明显,此时可能是保护缺血诱导神经元死亡的最佳时间。铁在髓鞘形成、神经递质合成与代谢、以及神经元树突发育中起重要作用。早期神经发育过程中铁的缺乏会影响海马功能的发育。反之,铁过载会产生氧自由基,从而损害细胞膜、蛋白和DNA。神经元对氧化损伤是十分敏感的。已有研究应用生化手段检测脑缺血后铁、铁蛋白和转铁蛋白的变化,但尚未见三者在短暂性脑缺血状态下细胞水平的变化。Dae Young Yoo等通过研究试图了解短暂性前脑缺血状态下,铁、铁蛋白和转铁蛋白在豚鼠海马CA1区的时间和细胞水平变化特征。研究发现,相对于假手术对照组,沙土鼠短暂性前脑缺血后12h,海马CA1区锥体细胞层铁反应明显上升。而为了代偿铁含量的增加,铁蛋白重链和转铁蛋白免疫反应在缺血后1d明显增强。研究为预防脑缺血后铁过载诱导的神经元死亡干预时间窗的选择提供了参考。文章发表于《中国神经再生研究(英文版)》杂志2016年第6期。
