摘要:

课题组前期研究发现,大鼠脑缺血再灌注损伤过程中脑组织Toll样受体3蛋白表达上调。鉴于此,实验以体外原代培养大鼠胎鼠皮质神经元建立缺糖缺氧模型,模拟脑缺血再灌注损伤过程,将化学合成的3对Toll样受体3基因小干扰RNA序列Toll样受体3-1280,1724,418转染入缺糖缺氧模型的皮质神经元中,以期抑制Toll样受体3蛋白表达的上调。Western-blot检测结果表明,3对Toll样受体3基因小干扰RNA转染后,皮质神经元Toll样受体3蛋白表达量均减少,尤以Toll样受体3-1724组抑制TLR3蛋白表达效果最为显著。结果证实体外缺糖缺氧条件下Toll样受体3-1724-小干扰RNA序列为抑制皮质神经元Toll样受体3表达的最优转染序列。
