Neural Regen Res:单唾液酸神经节苷酯1可能缓解丙泊酚联合瑞芬太尼对海马神经元的破坏作用
来自中国湖北医药学院麻醉学研究所陆江团队最新研究发现,单唾液酸神经节苷酯1可能缓解丙泊酚联合瑞芬太尼对神经干细胞的破坏作用,对未成熟中枢神经系统具有保护作用。
丙泊酚和瑞芬太尼相互作用可有效控制术中麻醉反应促进术后迅速苏醒,广泛用于全凭静脉麻醉,但有观点认为两者在神经发育方面存在不利影响,可能会损害未成熟的中枢神经系统。单唾液酸神经节苷酯1是神经节苷脂的主要亚型,在中枢神经系统中具有神经保护作用,可以抑制体内铅诱导的海马突触活动损伤和氧化损伤。
陆江等将从新生SD大鼠海马组织中分离的神经干细胞分别加入瑞芬太尼(5,10,20ng/mL)、丙泊酚(1.0,2.5,5.0μg/mL)和/或单唾液酸神经节苷酯1(12.5,25,50μg/mL),结果显示,单唾液酸神经节苷酯1能逆转丙泊酚联合瑞芬太尼干预后BrdU阳性细胞数量的减少,神经干细胞增殖和分化过程中凋亡细胞比例的增加以及神经干细胞分化过程中细胞内钙离子浓度的降低,但不影响神经干细胞分化过程中β-tubulin阳性细胞和神经胶质酸性蛋白阳性细胞的比例。这项成果撰写的文章发表在《中国神经再生研究(英文版)》杂志2017年6月6期。
