摘要:

对于神经系统疾病研究,小型猪无论是生理结构还是病理特征,均是最佳选择。但由于小型猪颅底部有一特征性生理结构——细动静脉血管组成的“异网”,这成为常规介入闭塞大脑中动脉方法建立小型猪缺血性脑卒中模型的瓶颈结构。实验探讨通过海藻酸钠微球堵塞小型猪脑基底部异网的方法建立缺血性脑卒中模型的可行性。共纳入3只巴马小型猪,在C型臂X光机监测下,经猪股动脉-主动脉弓-颈总动脉-咽升动脉-异网途径注入直径100-300 μm的新型栓塞材料海藻酸钠微球,并通过颈动脉造影、MRI检查、行为学观察和组织学分析对其进行评价。结果表明,模型猪单侧异网完全堵塞,基底节区供血障碍,右后肢站立不稳,口角流涎,MRI检查和苏木精-伊红染色可见模型猪大脑基底节区有明显梗死灶。说明海藻酸钠微球是一种较好的栓塞材料,利用该材料栓塞小型猪脑的特征性生理结构异网能够成功建立猪缺血性脑卒中模型,突破了猪不适用介入方法建立缺血性脑血管疾病模型的观念。
