摘要:

Ras/Raf/Mek/Erk信号级联通路的异常激活在脑胶质瘤的发生中起至关重要的作用,抑制该信号通路的过度激活,能有效抑制胶质瘤细胞增殖,促进其凋亡。那么神经干细胞是否也可通过抑制Ras/Raf/Mek/Erk信号通路发挥治疗脑胶质瘤的作用?实验将C6胶质瘤细胞悬液注射至大鼠颅脑制备脑胶质瘤模型,并将胎鼠神经干细胞植入脑胶质瘤模型大鼠脑内进行治疗。结果发现,脑胶质瘤模型大鼠瘤体组织Raf-1、Erk及抑凋亡蛋白Bcl-2表达显著增高,凋亡蛋白Caspase-3表达降低;神经干细胞移植后,脑胶质瘤模型大鼠瘤体组织Raf-1、Erk及Bcl-2蛋白表达显著下降,而Caspase-3蛋白表达显著升高。可见神经干细胞移植可通过抑制Ras/Raf/Mek/Erk信号通路,促进胶质瘤细胞凋亡。
