高压氧治疗缺氧缺血脑损伤新生大鼠脑室下区神经干细胞的增殖

Hyperbaric oxygen treatment promotes neural stem cell proliferation in the subventricular zone of neonatal rats with hypoxic-ischemic brain damage

摘要:

高压氧治疗新生儿缺血缺氧性脑病在临床上已应用多年,因作用机制尚不明确,对其疗效也存在争议。实验以新生7日龄SD乳鼠建立缺氧缺血性脑损伤模型,造模后6h应用高压氧连续干预7d,1h/d,观察乳鼠脑室下区神经干细胞的增殖。免疫组化染色显示,缺氧缺血性脑损伤后3d,乳鼠脑室下区BrdU+及nestin+细胞明显增多,损伤后5d达高峰。应用高压氧治疗的乳鼠脑室下区BrdU+及nestin+细胞在损伤后1d即开始增多,直至损伤后21d仍明显多于正常及缺氧缺血性脑损伤乳鼠。且苏木精-伊红染色显示给予高压氧干预的乳鼠脑组织病理损伤减轻,神经细胞变性和坏死减少。提示高压氧治疗可促进缺氧缺血性脑损伤乳鼠脑室下区神经干细胞增殖,从而减轻脑损伤。
