摘要:

小胶质细胞的激活通过释放多种促炎和神经毒性因子,对神经元的存活造成威胁。提取自毛茛科植物牡丹的干燥根皮的丹皮酚有抗氧化、抗炎和神经保护作用。为了解丹皮酚对炎症诱导的神经毒性和小胶质细胞激活的抑制效应。我们用脂多糖刺激大鼠大脑海马脑片和原代培养的小胶质细胞。在进行脂多糖刺激前,先以丹皮酚干预30min。结果发现:(1)在海马脑片中,丹皮酚抑制脂多糖诱导的海马细胞死亡和一氧化氮产物和白细胞介素1β的释放。(2)在小胶质细胞中,丹皮酚抑制脂多糖诱导的小胶质细胞释放一氧化氮、肿瘤坏死因子α和白细胞介素1β。结果说明丹皮酚能对炎症诱导的神经毒性产生抑制效应。
