电针干预抗抑郁:调节脑内多个信号靶点并提高脑源性神经营养因子水平

摘要:

电针比抗抑郁药物在改善抑郁行为方面作用更快,不良反应更少,但电针抗抑郁机制仍不明确。实验对以慢性不可预见性温和应激建立大鼠抑郁模型,然后进行电针治疗。电针刺激印堂和百会30min,频率2Hz,强度0.6mA,稀疏波,1次/d。结果显示,治疗2,4周时,电针治疗大鼠的水平和垂直活动增加,不动时间减少,海马中神经递质水平(5-羟色胺、谷氨酸和γ-氨基丁酸)及脑源性神经营养因子和脑源性神经营养因子相关蛋白(TrkB,蛋白激酶A,磷酸化环磷酸腺苷反应元件结合蛋白)水平上升,还发现蛋白激酶A和TrkB mRNA水平增加,钙调蛋白激酶II水平下降。提示电针通过调节环磷酸腺苷反应元件结合蛋白信号通路多个靶点,增加环磷酸腺苷反应元件结合蛋白磷酸化水平和脑源性神经营养因子水平,进而促进神经营养和神经再生,从而发挥抑郁作用。
