摘要:

鉴于神经细胞对氧化损伤的易感性及硫氧还蛋白还原酶1在中枢神经系统分布的广泛性及抗氧化特性,我们推测硫氧还蛋白还原酶1可能参与了帕金森病的发病。为此,实验采用1-甲基-4-苯基-1,2,3,6-四氢吡啶腹腔注射制备帕金森病小鼠模型。以实时RT-PCR、western blot、比色法检测显示,帕金森病模型小鼠中脑硫氧还蛋白还原酶1 mRNA和蛋白的表达、硫氧还蛋白还原酶的活性均明显降低。免疫组织化学染色显示,帕金森病模型小鼠中脑黑质致密区硫氧还蛋白还原酶1免疫阳性神经元的数量显著减少。结果说明,硫氧还蛋白还原酶1蛋白在帕金森病小鼠黑质致密区表达显著下调,可能与帕金森病的发病有关。
